桃种皮细胞的超微结构观察及生长素结合蛋白ABP1亚细胞定位

摘  要  为探究桃种皮细胞的超微结构特征以及ABP1在种皮中的亚细胞水平分布特点,本试验以“大久保”桃种皮为实验材料,利用透射电子显微镜观察了种皮细胞超微结构的变化,采用免疫胶体金标记技术对内源ABP1进行定位分析。结果显示:种皮细胞结构具有明显的发育时期特异性。ABP1信号主要出现在细胞核、内质网、质膜、细胞壁、线粒体和液泡上,而较强的信号分布于内质网和质膜上。本试验结果为ABP1在桃种皮细胞中的具体分布及其变化提供直观的形态学证据,为进一步研究其在果实发育过程中的作用机制奠定了理论基础。

Observation of cellular ultrastructure and subcellular localization of ABP1 in seed coat of peach fruit

Abstract  To explore the ultrastructural characteristics and the subcellular distribution level of ABP1 in the seed coat during peach fruit development, the peach ‘dajiubao’ was used as raw material to observe the ultrastructural changes in the seed coat by electron microscope, to analyze the location of endogenous ABP1 by immunogold labeling electron microscopy. The results show that significant changes appeare in the structure of the seed coat cells with the development of peach fruit, and ABP1 signals mainly distribute in the nucleus, endoplasmic reticulum, plasma membrane, cell wall and vacuoles. The stronger signal appeares on the endoplasmic reticulum and the plasma membrane. This study provides an intuitive morphological evidence for the specific distribution and changes of ABP1 in the seed coat cells, and lays a good foundation for the further study on the mechanism of ABP1 in the process of peach fruit development.
